Fusion of the inner and outer nuclear membranes during pore complex assembly

The assembly of nuclear pore complexes (NPCs) during interphase begins with the recruitment of nucleoporins to the nuclear side of the inner nuclear membrane. This recruitment induces membrane curvature, ultimately leading to fusion of the inner and outer nuclear membranes and insertion of NPCs at the fusion sites. In this proposal, we aim to elucidate how the integral membrane proteins Apq12, Brl1, and Brr6 mediate this fusion process during NPC assembly, using genetic, cell biological, and biochemical approaches. By doing so, we address a fundamental question in cell biology with broad implications for cell division, aging, and cancer development.
